Myoglobin is a cytosolic oxygen-binding protein responsible for the storage and diffusion of oxygen within myocytes. Expression of myoglobin is highest in skeletal and cardiac muscle. Myoglobin is well accepted as an O2-storage protein in muscle, capable of releasing O2 during periods of hypoxia or anoxia. In combination with other striated muscle markers such as vimentin and myogenin, myoglobin is helpful in the identification of rhabdomyosarcoma and tumors with skeletal muscle differentiation. Reportedly, myoglobin is expressed on epithelial cancer cells due to changed metabolic and environmental conditions. This monoclonal antibody is specific to estrogen receptor alpha (ER alpha) and shows minimal cross-reaction with other members of the family. This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Calretinin (also known as Calbindin 2) is a member of the troponin C superfamily. Proteins in this family have 6 EF-hand domains which bind to calcium. Calretinin is a vitamin D-dependent calcium-binding protein involved in calcium signaling. It is present in subsets of neurons throughout the brain and spinal chord, including sensory ganglia. Antibodies to calretinin are useful in differentiating mesothelioma from adenocarcinomas of the lung. This antibody recognizes a sialoglycoprotein of 27-32 kDa, identified as CD99, or MIC2 gene product, or E2 antigen. MIC2 gene is located in the pseudo-autosomal region of the human X and Y chromosome. MIC2 gene encodes two distinct proteins, which are produced by alternative splicing of the CD99 gene transcript and are identified as bands of 30 and 32 kDa (p30/32).Although its function is not fully understood, CD99 is implicated in various cellular processes including homotypic aggregation of T cells, upregulation of T cell receptor and MHS molecules, apoptosis of immature thymocytes and leukocyte diapedesis. CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. Most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells express this antigen. Mature granulocytes express very little or no CD99. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Napsin is a pepsin-like aspartic proteinase connected with maturation of surfactant protein B. There are two closely related napsins, napsin A and napsin B. Napsin A is expressed as a single chain protein. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. Napsin A is expressed in type II pneumocytes and in adenocarcinomas of lung. The high specificity expression of napsin A in adenocarcinomas of lung is useful to distinguish primary lung adenocarcinomas from adenocarcinomas of other organs. This antibody recognizes a protein of 62 kDa, which is identified as AKT1. The serine/threonine kinase Akt family contains several members, including Akt1 (also designated PKB or RacPK), Akt2 (also designated PKBbeta or RacPK-beta) and Akt3 (also designated PKB-gamma or thymoma viral proto-oncogene 3), which exhibit sequence homology with the protein kinase A and C families and are encoded by the c-Akt proto-oncogene. This MAb recognizes an epitope within the 74-182 C-terminal sequence (11kD peptide fragment) of human serum Cellular Retinol Binding Protein 1 (CRBP 1), a single-chain glycoprotein belonging to the superfamily of hydrophobic molecule transporter proteins, which is responsible for transport of retinol (vitamin A1) from the liver to peripheral target tissues, like the eye, where it mediates the cellular uptake.
